The patient's ABO type and rhesus status must be known. A sample of the potential donor red cells is then incubated with a sample of patient serum in a test tube and checked for agglutination.

Grouping, antibody screening, and cross-matching take about an hour and a half which is usually rapid enough to avoid the use of universal donor blood - Group O negative.
