Corynebacteria grow aerobically on nutrient agar at 37 degrees centigrade. However, they exhibit better growth on blood or serum agar. With brain-heart infusion agar, colonies form within 24 hours and are convex, circular and cream-coloured.
Loeffler's serum is the special medium used for the isolation of C. diphtheriae.
